Theodora Tzakri ist in Pella geboren und aufgewachsen. Sie ist verheiratet und hat eine Tochter. Sie hat ihr Studium an der Rechtswissenschaftlichen Fakultät der Aristoteles-Universität Thessaloniki (AUTh) abgeschlossen und einen Master-Abschluss in Öffentlichem Recht und Politikwissenschaft der Rechtswissenschaftlichen Fakultät der Demokrit-Universität Thrakien erworben.
Sie spricht Englisch und Deutsch. Seit 1996 ist sie als Rechtsanwältin tätig. Von 1998 bis 2004 war sie gewählte Regionalvertreterin in Pella und hatte das Amt der Vizepräfektin für Bildung inne.
Seit 2004 ist sie Mitglied des griechischen Parlaments. Sie war stellvertretende Ministerin für Inneres (2009–2011), stellvertretende Ministerin für Industrie (2015–2017) und Sekretärin der SYRIZA-Fraktion (2023–2024). Seit 2025 ist sie Vizepräsidentin der EDP.
