Theodora Tzakri è nata e cresciuta a Pella. È sposata e ha una figlia. Si è laureata alla Facoltà di Giurisprudenza dell'Università Aristotele di Salonicco (AUTh) e ha conseguito un master in Diritto pubblico e Scienze politiche presso la Facoltà di Giurisprudenza dell'Università Democrito di Tracia.
Parla inglese e tedesco. Esercita la professione legale dal 1996. Dal 1998 al 2004 è stata rappresentante regionale eletta a Pella e ha ricoperto la carica di viceprefetto per l'istruzione.
È membro del Parlamento ellenico dal 2004. Ha ricoperto la carica di viceministro degli Affari interni (2009-2011), viceministro dell'Industria (2015-2017) e segretario del gruppo parlamentare SYRIZA (2023-2024). Dal 2025 è vicepresidente del PDE.
